Evaluation of genus Thermoanaerobacter’s alcohol dehydrogenase specificities

Several of the alcohol dehydrogenases (ADHs) of species within the genus Thermoanaerobacter are catalysts for secondary hydroxyl groups with the secondary ADH (SADH) produced by T. pseudoethanolicus being particularly well described in the literature. Unfortunately, this species SADH has a limited range of substrates which it can accept within its binding pocket without modification. This work examines the substrate specificity of the NAD and NADP-linked ADHs within the genus, as well as the closely related genus of Caldanaerobacter, and several strains isolated from hot springs in Iceland. Several of the strains evaluated have different specificities than those previously published. Work on the use of both partially purified and whole-cell systems for the resolution of useful alcohols and diols will be examined with an emphasis on historically difficult to resolve molecules such as 1,2-alkanediols.
