MITF (microphthalmia associated transcription factor) is the master regulator of melanocytes. In melanoma, MITF acts as a molecular switch that determines whether melanoma cells differentiate, proliferate or become quiescent migratory cells able to form metastasis. Currently, it is not understood how MITF can mediate all these effects. In this study we use ChIP-seq and RNA-seq to characterize the MITF transcription factor network involving MITF, IRF4, and TFEB, determine how they cooperate to regulate a range of genes important in melanomagenesis and characterize how different levels of MITF activity affect progression of melanoma.
