Introduction: The Blood bank has developed methods that use lysate from expired pathogen inactivated platelet concentrates (PIPL) as supplements for expansion and osteogenic differentiation of mesenchymal stem cells (MSC). Utilization of such lysates eliminates the use of animal serum on MSC cell culture. Objective: The aim of this project was to compare the use of PIPL and freeze-dried PIPL (L-PIPL) on expansion and osteogenic differentiation of MSC. Methods: Bone marrow derived MSC were expanded in media supplemented with either PIPL or L-PIPL and evaluated using XTT assay and population doubling assay. Osteogenic differentiation was analysed with alizarin red quantification and alkaline phosphatase activity assay. Results and conclusion: Preliminary data indicates that both PIPL and L-PIPL are suitable for expansion and osteogenic differentiation of bone marrow derived MSC. More detailed analysis is underway that includes gene expression analysis of osteogenic markers as well as detailed characterization of PIPL and L-PIPL using cytokine and protein analysis.
